Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.159A>C (p.Glu53Asp), citing Ambry Variant Classification Scheme 2023: The p.E53D variant (also known as c.159A>C), located in coding exon 2 of the DICER1 gene, results from an A to C substitution at nucleotide position 159. The glutamic acid at codon 53 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
