It has not been published as pathogenic or been reported as benign to our knowledge. Additionally, the S738C variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S738C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species and in silico analysis predicts this variant is probably damaging to the protein structure/function. Furthermore, likely pathogenic variants at the same residue (S738R, S738T), and pathogenic/likely pathogenic missense variants in nearby residues (I736L, I736V, I736T, I736M, G741R, G741W, G741E) have been reported at GeneDx and/or reported in the Human Gene Mutation Database in association with HCM (Stenson et al., 2014), supporting the functional importance of this residue and region of the protein.
