NM_016222.4(DDX41):c.4G>A (p.Glu2Lys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2K variant (also known as c.4G>A), located in coding exon 1 of the DDX41 gene, results from a G to A substitution at nucleotide position 4. The glutamic acid at codon 2 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
