NM_001386795.1(DTNA):c.2228G>A (p.Arg743Gln) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the DTNA gene. The R716Q variant has not been published as pathogenic or been reported as benign to our knowledge. Additionally, this variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R716Q variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. However, while this substitution occurs at a position that is conserved in mammals, Glutamine is the wild-type residue at this position in one mammalian species. Furthermore, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
