NM_020699.4(GATAD2B):c.1414del (p.Glu472fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.1414delG variant in the GATAD2B gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The 1414delG variant causes a frameshift starting with codon Glutamic acid 472, changes this amino acid to an Asparagine residue, and creates a premature Stop codon at position 32 of the new reading frame, denoted p.E472NfsX32. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The 1414delG variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret c.1414delG as a pathogenic variant
