NM_001103.4(ACTN2):c.2539G>A (p.Ala847Thr) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the ACTN2 gene (transcript NM_001103.4) at coding-DNA position 2539, where G is replaced by A; at the protein level this means replaces alanine at residue 847 with threonine — a missense variant. Submitter rationale: The A847T variant has not been published as pathogenic or been reported as benign to our knowledge. It is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The A847T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. In addition, this substitution occurs at a position that is conserved through mammals. Nonetheless, in silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function.
