Uncertain significance — the classification assigned by GeneDx to NM_002887.4(RARS1):c.1568T>A (p.Met523Lys), citing GeneDx Variant Classification (06012015). This variant lies in the RARS1 gene (transcript NM_002887.4) at coding-DNA position 1568, where T is replaced by A; at the protein level this means replaces methionine at residue 523 with lysine — a missense variant. Submitter rationale: The M523K variant in the RARS gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The M523K variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The M523K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position [that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret M523K as a variant of uncertain significance.
