Pathogenic for Marfan Syndrome/Loeys-Dietz Syndrome/Familial Thoracic Aortic Aneurysms and Dissections — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000138.5(FBN1):c.4505G>A (p.Cys1502Tyr), citing LabCorp Variant Classification Summary - May 2015: Variant summary: FBN1 c.4505G>A (p.Cys1502Tyr) results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251106 control chromosomes. c.4505G>A has been observed in individual(s) affected with Marfan Syndrome (examples: Ganesh_2006, Stheneur_2009, internal data). In at least one individual the variant was observed to be de novo. These data indicate that the variant is likely to be associated with disease. Based on the evidence outlined above, the variant was classified as pathogenic.
