NM_001429.4(EP300):c.3412dup (p.Val1138fs) was classified as Pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): The c.3412dupG pathogenic variant in the EP300 gene causes a frameshift starting with codon Valine 1138, changes this amino acid to a Glycine residue and creates a premature Stop codon at position 10 of the new reading frame, denoted p.Val1138GlyfsX10. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. It is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server).
