Pathogenic — the classification assigned by GeneDx to NM_001367721.1(CASK):c.2191GTA[1] (p.Val732del), citing GeneDx Variant Classification (06012015): The c.2179_2181delGTA variant in the CASK gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The c.2179_2181delGTA variant causes an inframe deletion of codon Valine 727, denoted p.Val727del. The c.2179_2181delGTA variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). This deletion occurs at a position that is conserved across species. We interpret c.2179_2181delGTA as a pathogenic variant.
