NM_006073.4(TRDN):c.553A>G (p.Thr185Ala) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the TRDN gene (transcript NM_006073.4) at coding-DNA position 553, where A is replaced by G; at the protein level this means replaces threonine at residue 185 with alanine — a missense variant. Submitter rationale: A variant of uncertain significance has been identified in the TRDN gene. The T185A variant has not been published as pathogenic or been reported as benign to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The T185A variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. However, this substitution occurs at a position that is not conserved across species and where alanine is present as the wild type in at least one species. In silico analysis suggests that this variant likely does not alter the protein structure/function.
