Other variant(s) that disrupt this residue have been observed in individuals with FBN1-related conditions (PMID: 26747767, Invitae), which suggests that this may be a clinically significant amino acid residue. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This variant has been observed in individuals affected with Marfan syndrome or FBN1-related conditions (PMID: 24793577, Invitae). ClinVar contains an entry for this variant (Variation ID: 42343). This variant is not present in population databases (ExAC no frequency). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces glutamic acid with alanine at codon 1283 of the FBN1 protein (p.Glu1283Ala). The glutamic acid residue is highly conserved and there is a moderate physicochemical difference between glutamic acid and alanine.
