NM_000540.3(RYR1):c.3751C>A (p.His1251Asn) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the RYR1 gene (transcript NM_000540.3) at coding-DNA position 3751, where C is replaced by A; at the protein level this means replaces histidine at residue 1251 with asparagine — a missense variant. Submitter rationale: The H1251N variant in the RYR1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The H1251N variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The H1251N variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret H1251N as a variant of uncertain significance.
