This variant lies in the COL5A1 gene (transcript NM_000093.5) at coding-DNA position 4796 through coding-DNA position 4798, replacing the reference sequence with TGG. Submitter rationale: The c.4796_4798delAGAinsTGG variant (also known as p.E1599_N1600delinsVD), located in coding exon 62 of the COL5A1 gene, results from an in-frame deletion of AGA and insertion of TGG at nucleotide positions 4796 to 4798. This results in the substitution of the glutamic acid and asparagine residues for valine and aspartic acid residues at codons 1599 and 1600, respectively. These amino acid positions are not well conserved in available vertebrate species. In addition, this variant is predicted to be neutral by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Based on the available evidence, the clinical significance of this variant remains unclear.
