NM_000089.4(COL1A2):c.2452C>G (p.Leu818Val) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L818V variant (also known as c.2452C>G), located in coding exon 40 of the COL1A2 gene, results from a C to G substitution at nucleotide position 2452. The leucine at codon 818 is replaced by valine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
