Uncertain significance — the classification assigned by GeneDx to NM_021939.4(FKBP10):c.506G>C (p.Arg169Pro), citing GeneDx Variant Classification (06012015): The R169P variant in the FKBP10 gene has not been reported previously as a pathogenic variant, noras a benign variant, to our knowledge. The R169P variant is not observed at a significant frequency inlarge population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome VariantServer). The R169P variant is a non-conservative amino acid substitution, which occurs at a positionthat is conserved across species. In silico analysis predicts this variant is probably damaging to theprotein structure/function. We interpret R169P as a variant of uncertain significance.
