NM_138422.4(ADAT3):c.1093C>G (p.Pro365Ala) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015). This variant lies in the ADAT3 gene (transcript NM_138422.4) at coding-DNA position 1093, where C is replaced by G; at the protein level this means replaces proline at residue 365 with alanine — a missense variant. Submitter rationale: The P365A variant in the ADAT3 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. This variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The P365A variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. This substitution occurs at a position that is not conserved. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret P365A as a variant of uncertain significance.
