Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000722.4(CACNA2D1):c.897G>C (p.Gln299His), citing Ambry Variant Classification Scheme 2023: The p.Q299H variant (also known as c.897G>C), located in coding exon 11 of the CACNA2D1 gene, results from a G to C substitution at nucleotide position 897. The glutamine at codon 299 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and histidine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
