Uncertain significance — the classification assigned by GeneDx to NM_201384.3(PLEC):c.9823C>T (p.Arg3275Cys), citing GeneDx Variant Classification (06012015): The R3302C variant in the PLEC gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The R3302C variant is not observed at a significant frequency in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The R3302C variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Arginine are tolerated across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. We interpret R3302C as a variant of uncertain significance.
