This variant lies in the DMD gene (transcript NM_004006.3) at coding-DNA position 5149, where C is replaced by T; at the protein level this means replaces leucine at residue 1717 with phenylalanine — a missense variant. Submitter rationale: The L1717F variant in the DMD gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The L1717F variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L1717F variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. This substitution occurs at a position where amino acids with similar properties to Leucine are tolerated across species.
