Pathogenic — the classification assigned by GeneDx to NM_005249.5(FOXG1):c.1058dup (p.Asn353fs), citing GeneDx Variant Classification (06012015): The c.1058dupA pathogenic variant in the FOXG1 gene causes a frameshift starting with codon Asparagine 353, changes this amino acid to a Lysine residue and creates a premature Stop codon at position 102 of the new reading frame, denoted p.Asn353LysfsX102. This pathogenic variant is predicted to cause loss of normal protein function through protein truncation, as the last 137 amino acids are replaced with 101 incorrect amino acids. Furthermore, the c.1058dupA variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server).
