Submitter rationale: The c.222_223insGAC variant in the KIF1A gene has not been reported previously as a pathogenic variant nor as a benign variant, to our knowledge. The c.222_223insGAC variant causes an in-frame insertion of an Aspartic Acid residue between codons Tyrosine 74 and Arginine 75, denoted p.Tyr74_Arg75insAsp. The c.222_223insGAC variant is located in the kinesin motor domain, a functionally important region of the protein (Hamdan et al., 2011). The c.222_223insGAC variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. We interpret c.222_223insGAC as a pathogenic variant
