This variant lies in the CHEK2 gene (transcript NM_007194.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1542, where G is replaced by T; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.1542+1G>T intronic variant results from a G to T substitution one nucleotide after coding exon 13 of the CHEK2 gene. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing. This nucleotide position is highly conserved in available vertebrate species.
