NM_000492.4(CFTR):c.3812G>A (p.Gly1271Asp) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1271D variant (also known as c.3812G>A), located in coding exon 23 of the CFTR gene, results from a G to A substitution at nucleotide position 3812. The glycine at codon 1271 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
