Uncertain Significance for Lynch syndrome — the classification assigned by All of Us Research Program, National Institutes of Health to NM_000251.3(MSH2):c.-13AGG[1], citing ACMG Guidelines, 2015: This variant causes the deletion of 3 nucleotides in the 5' untranslated region in the MSH2 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with hereditary cancer in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
