NM_000075.4(CDK4):c.405C>G (p.Cys135Trp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C135W variant (also known as c.405C>G), located in coding exon 3 of the CDK4 gene, results from a C to G substitution at nucleotide position 405. The cysteine at codon 135 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
