This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 5593, deleting one base; at the protein level this means shifts the reading frame starting at isoleucine residue 1865, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: This deletion of one nucleotide in BRCA2 is denoted c.5593delA at the cDNA level and p.Ile1865TyrfsX9 (I1865YfsX9) at the protein level. Using alternate nomenclature, this variant would be defined as BRCA2 5821delA. The normal sequence, with the base that is deleted in braces, is AGAC[A]TATT. The deletion causes a frameshift which changes an Isoleucine to a Tyrosine at codon 1865, and creates a premature stop codon at position 9 of the new reading frame. Although this variant has not, to our knowledge, been reported in the literature, it is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay.
