Pathogenic — the classification assigned by GeneDx to NM_004656.4(BAP1):c.1254T>A (p.Tyr418Ter), citing GeneDx Variant Classification (06012015): This variant is denoted BAP1 c.1254T>A at the cDNA level and p.Tyr418Ter (Y418X) at the protein level. The substitution creates a nonsense variant, which changes a Tyrosine to a premature stop codon (TAT>TAA), and is predicted to cause loss of normal protein function through either protein truncation or nonsense-mediated mRNA decay. Although this variant has not, to our knowledge, been reported in the literature, it is considered pathogenic.
