Likely pathogenic — the classification assigned by GeneDx to NM_152416.4(NDUFAF6):c.600del (p.Asp201fs), citing GeneDx Variant Classification (06012015): The c.600delA variant in the NDUFAF6 gene causes a frameshift starting with codon Aspartic acid 201, changes this amino acid to an Isoleucine residue and creates a premature Stop codon at position 17 of the new reading frame, denoted p.Asp201IlefsX17. This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. Although this variant has not been previously reported to our knowledge, it is likely to be a pathogenic variant.
