Uncertain significance — the classification assigned by GeneDx to NM_138477.4(CDAN1):c.1567G>A (p.Val523Ile), citing GeneDx Variant Classification (06012015): The V523I variant in the CDAN1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The V523I variant is observed in 49/276702 (0.018%) alleles in large population cohorts (Lek et al., 2016). The V523I variant is a conservative amino acid substitution, which is not likely to impact secondary protein structure as these residues share similar properties. In-silico analyses, including protein predictors and evolutionary conservation, support that this variant does not alter protein structure/function. We interpret V523I as a variant of uncertain significance.
