Pathogenic for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_181426.2(CCDC39):c.1893_1896delinsTT (p.Arg631_Leu632insTer), citing Ambry Variant Classification Scheme 2023: The c.1893_1896delGCTAinsTT pathogenic mutation, located in coding exon 14 of the CCDC39 gene, results from the deletion of 4 nucleotides and insertion of two nucleotides causing a translational frameshift with a predicted alternate stop codon (p.L632*). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
