NM_000530.8(MPZ):c.149_151dup (p.Cys50dup) was classified as Likely pathogenic by GeneDx, citing GeneDx Variant Classification (06012015): A novel c.149_151dupGCT variant that is likely pathogenic has been identified in the MPZ gene. The c.149_151dupGCT variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. It was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The c.149_151dupGCT variant results in an in-frame duplication of a single Cysteine residue, denoted p.Cys50dup. This variant occurs at a conserved position predicted to be within the extracellular immunoglobulin-like domain of the MPZ protein (Lee et al., 2010). In silico analysis predicts this variant is probably damaging to the protein structure/function. Therefore, this variant is likely pathogenic; however, the possibility that it is benign cannot be excluded.
