Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001232.4(CASQ2):c.33T>G (p.Ile11Met), citing Ambry Variant Classification Scheme 2023: The p.I11M variant (also known as c.33T>G), located in coding exon 1 of the CASQ2 gene, results from a T to G substitution at nucleotide position 33. The isoleucine at codon 11 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
