NM_000722.4(CACNA2D1):c.2920G>T (p.Asp974Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D974Y variant (also known as c.2920G>T), located in coding exon 36 of the CACNA2D1 gene, results from a G to T substitution at nucleotide position 2920. The aspartic acid at codon 974 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
