Submitter rationale: The p.S2616A variant (also known as c.7846T>G), located in coding exon 16 of the BRCA2 gene, results from a T to G substitution at nucleotide position 7846. The serine at codon 2616 is replaced by alanine, an amino acid with similar properties. Two saturation genome editing-based studies, including a haploid cell-survival assay and a humanized mouse embryonic stem cell line assay of drug response and survival, demonstrate that this nucleotide substitution may be functional; however, additional evidence is needed to confirm these findings (Huang H et al.
