Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.4798T>A (p.Leu1600Met), citing Ambry Variant Classification Scheme 2023: The p.L1600M variant (also known as c.4798T>A), located in coding exon 14 of the BRCA1 gene, results from a T to A substitution at nucleotide position 4798. The leucine at codon 1600 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
