Likely pathogenic — the classification assigned by GeneDx to NM_006086.4(TUBB3):c.1081C>A (p.Leu361Ile), citing GeneDx Variant Classification (06012015): The L361I variant in the TUBB3 gene has not been reported previously as a pathogenic variant, noras a benign variant, to our knowledge. The L361I variant is not observed in large population cohorts(Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The L361I variantis a conservative amino acid substitution, which is not likely to impact secondary protein structure asthese residues share similar properties. This substitution occurs at a position that is conserved acrossspecies. In silico analysis is inconsistent in its predictions as to whether or not the variant is damagingto the protein structure/function. The L361I variant is a strong candidate for a pathogenic variant; however the possibility itmay be a rare benign variant cannot be excluded.
