This variant lies in the POLD1 gene (transcript NM_002691.4) at the canonical splice donor site of the intron immediately after 2 bases upstream of the translation start (5' untranslated region), where G is replaced by C; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: The c.-2+1G>C intronic variant is located in the 5' untranslated region (5&rsquo; UTR) of the POLD1 gene. This intronic variant results from a G to C substitution one nucleotide after the first translated codon. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. Alterations that disrupt the canonical splice site are expected to cause aberrant splicing, resulting in an abnormal protein or a transcript that is subject to nonsense-mediated mRNA decay.
