Uncertain significance — the classification assigned by GeneDx to NM_001278116.2(L1CAM):c.2249A>C (p.Tyr750Ser), citing GeneDx Variant Classification (06012015). This variant lies in the L1CAM gene (transcript NM_001278116.2) at coding-DNA position 2249, where A is replaced by C; at the protein level this means replaces tyrosine at residue 750 with serine — a missense variant. Submitter rationale: The Y750S variant in the L1CAM gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The Y750S variant is not observed in large population cohorts (Lek et al., 2016). The Y750S variant is a semi-conservative amino acid substitution, which may impact secondary protein structure as these residues differ in some properties. In-silico analyses, including protein predictors and evolutionary conservation, support a deleterious effect. We interpret Y750S as a variant of uncertain significance.
