This variant lies in the DDX41 gene (transcript NM_016222.4) at coding-DNA position 91, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 31 with lysine — a missense variant. Submitter rationale: The E31K variant in the DDX41 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The E31K variant was not observed in approximately 6500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The E31K variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position where amino acids with similar properties to Glutamic Acid are tolerated across species. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret E31K as a variant of uncertain significance.
