Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to Single allele, citing Ambry Variant Classification Scheme 2023: The p.Y2423H variant (also known as c.7267T>C), located in coding exon 2 of the ZNF469 gene, results from a T to C substitution at nucleotide position 7267. The tyrosine at codon 2423 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
