Single allele was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R3447L variant (also known as c.10340G>T), located in coding exon 2 of the ZNF469 gene, results from a G to T substitution at nucleotide position 10340. The arginine at codon 3447 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
