Uncertain significance — the classification assigned by GeneDx to NM_001323289.2(CDKL5):c.1060C>T (p.Pro354Ser), citing GeneDx Variant Classification (06012015): A variant of uncertain significance has been identified in the CDKL5 gene. The P354S variant hasnot been published as a pathogenic variant, nor has it been reported as a benign variant to ourknowledge. It was not observed in approximately 6,500 individuals of European and AfricanAmerican ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benignvariant in these populations. The P354S variant is a non-conservative amino acid substitution,which is likely to impact secondary protein structure as these residues differ in polarity, charge,size and/or other properties. However, this substitution occurs at a position that is not conserved.In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging tothe protein structure/function. Therefore, based on the currently available information, it is unclearwhether this variant is a pathogenic variant or a rare benign variant.
