Pathogenic for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001174096.2(ZEB1):c.787_789delinsTAA (p.His263Ter), citing Ambry Variant Classification Scheme 2023: The c.784_786delCACinsTAA (p.H262*) alteration, located in exon 6 (coding exon 6) of the ZEB1 gene, consists of an in-frame substitution of 3 nucleotides from position 784 to 786. This changes the amino acid from a histidine to a stop codon at amino acid position 262. This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Based on the available evidence, this alteration is classified as pathogenic.
