This variant lies in the MAP2K2 gene (transcript NM_030662.4) at 10 bases into the intron immediately after coding-DNA position 919 through 12 bases into the intron immediately after coding-DNA position 919, replacing the reference sequence with AAG. Submitter rationale: Variant summary: MAP2K2 c.919+10_919+12delinsAAG alters a nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. 4/4 computational tools predict no significant impact on normal splicing. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 211110 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.919+10_919+12delinsAAG in individuals affected with Noonan Syndrome and Related Conditions and no experimental evidence demonstrating its impact on protein function have been reported.
