This variant lies in the CNGB1 gene (transcript NM_001297.5) at coding-DNA position 3010, where G is replaced by T; at the protein level this means replaces alanine at residue 1004 with serine — a missense variant. Submitter rationale: The A1004S variant in the CNGB1 gene has not been reported previously as a pathogenic variant, nor as a benign variant, to our knowledge. The A1004S variant was not observed in approximately 6200 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The A1004S variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved in mammals. In silico analysis is inconsistent in its predictions as to whether or not the variant is damaging to the protein structure/function. We interpret A1004S as a variant of uncertain significance.
