Pathogenic — the classification assigned by GeneDx to NM_000444.6(PHEX):c.985dup (p.His329fs), citing GeneDx Variant Classification (06012015): The c.985dupC pathogenic variant in the PHEX gene has been reported previously in association with X-linked hypophosphatemic rickets (Rowe et al., 1997). The duplication causes a frameshift starting with codon Histidine 329 changes this amino acid to a Proline residue and creates a premature Stop codon at position 20 of the new reading frame, denoted p.His329ProfsX20. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay.
