Pathogenic — the classification assigned by GeneDx to NM_001042492.3(NF1):c.2545G>T (p.Gly849Ter), citing GeneDx Variant Classification (06012015): The G849X nonsense variant in the NF1 gene is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). Although this pathogenic variant has not been reported previously to our knowledge, we consider it to be pathogenic.
