Pathogenic — the classification assigned by GeneDx to NM_001009944.3(PKD1):c.1687C>T (p.Gln563Ter), citing GeneDx Variant Classification (06012015): The Q563X variant in the PKD1 gene has been reported previously in a family with polycystic kidney disease (McCluskey et al., 2002). This variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay. The Q563X variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). We interpret Q563X as a pathogenic variant.
