Submitter rationale: The C204Y variant in the FZD4 gene has been reported in association with familial exudative vitroretinopathy (Nikopoulos et al. 2010). In vitro functional studies demonstrated that the presence of the C204Y variant results in the protein being exclusively retained within the endoplasmic reticulum (Milhem et al. 2014). C204Y was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The C204Y variant is a non-conservative amino acid substitution that occurs at a position that is conserved across species. Therefore, we interpret C204Y as a pathogenic variant.
